چکیده

The human body is constantly challenged by a variety of commensal and pathogenic microorganisms that trigger the immune system. Central in the first line of defence is the patternrecognition receptor (PRR)-induced stimulation of the NFkB pathway, leading to NFkB activation. The subsequent production of pro-inflammatory cytokines and/or antimicrobial peptides results in recruitment of professional phagocytes and bacterial clearance. To overcome this, bacteria have developed mechanisms for targeted interference in every single step in the PRR–NFkB pathway to dampen host inflammatory responses. This review aims to briefly overview the PRR–NFkB pathway in relation to the immune response and give examples of the diverse bacterial evasion mechanisms including changes in the bacterial surface, decoy production and injection of effector molecules. Targeted regulation of inflammatory responses is needed and bacterial molecules developed for immune evasion could provide future anti-inflammatory agents.

Anatomy of a negative feedback loop: the case of IkBa

The magnitude, duration and oscillation of cellular signalling pathway responses are often limited by negative feedback loops, defined as an ‘activator-induced inhibitor’ regulatory motif. Within the NFkB signalling pathway, a key negative feedback regulator is IkBa. We show here that, contrary to current understanding, NFkB-inducible expression is not sufficient for providing effective negativ...

Agonistic and Antagonistic Roles for TNIK and MINK in Non-Canonical and Canonical Wnt Signalling

Wnt signalling is a key regulatory factor in animal development and homeostasis and plays an important role in the establishment and progression of cancer. Wnt signals are predominantly transduced via the Frizzled family of serpentine receptors to two distinct pathways, the canonical ß-catenin pathway and a non-canonical pathway controlling planar cell polarity and convergent extension. Interfe...

Attenuation of NFkB Activation Augments Alveolar Transport Proteins Expression and Activity under Hypoxia

Epithelial sodium channels (ENaCs) controls vectorial Na across epithelia thus playing a central role in all aspects of fluid clearance in the body. The aim of this study was to investigate in vitro time dependant model of hypoxia induced reduction in vectorial Na and fluid transport across the alveolar epithelium. AEC exposed to 3% O2, for different time periods (1 h, 3h, 6h, 12h, 24h and 48 h...

IkBb enhances the generation of the low-affinity NFkB/RelA homodimer

The NFkB family of dimeric transcription factors regulate inflammatory and immune responses. While the dynamic control of NFkB dimer activity via the IkB–NFkB signalling module is well understood, there is little information on how specific dimer repertoires are generated from Rel family polypeptides. Here we report the iterative construction—guided by in vitro and in vivo experimentation—of a ...
